Primary bone marrow mononuclear cells (a.k.a. BM MNCs or BMNCs) are a heterogeneous population that includes hematopoietic lineage cells such as lymphocytes, monocytes, stem cells, and progenitor cells as well as mesenchymal stromal cells. Mononuclear cells are isolated from human adult bone marrow using density gradient separation. Heparin is added to the bone marrow upon collection as an anticoagulant.Cells were obtained using Institutional Review Board (IRB) approved consent forms and protocols.Certain products are only available in select territories. The PathHunter EDG7 Activated GPCR internalization cell line is a stable, engineered cell line for use in studying drug candidates (small molecules or biologics) affecting GPCR activation and internalization. This cell line uses EFC technology to measure ligand-induced internalization of EDG7 via detection of localization to the early endosome. The included cell line overexpresses EA-tagged β-Arrestin-2 as well as a ProLink-tagged early endosome protein and untagged EDG7. Upon ligand-induced receptor activation, β-Arrestin-2 is recruited to the activated GPCR and the resulting EDG7-arrestin complex is internalized to the early endosome, leading to production of EFC signal. Upon activation of TRHR, the resulting calcium activity can be fluorescently assayed.
